The couple welcomed a daughter, Elsie Marigold, last month, but only confirmed the news this week.
In a statement to E! News, the couple said: "We are beyond thrilled to announce the arrival of our daughter, Elsie Marigold, on September 13. A little sister for Ella Betsi.
"We both feel very grateful and very blessed."
It is understood the baby was due on 25 September, but arrived early in Los Angeles, where the British couple now live.
The pair first met in 2000 on the film '102 Dalmatians'.
They married in Mexico in 2007 and are already parents to four-year-old Ella Betsi.
Gruffudd, 39, previously spoke about fatherhood to the Daily Mail.
He said: "We'd wanted a baby for a while and after it didn't happen for us naturally we said, 'Let's take what science has to offer.' Luckily for us, it worked the first time so it wasn't a long, hard struggle."
He added that his first daughter is "very, very strong", and joked: "She's like her mother. I've got my work cut out for me at home."
