A spokesperson for the couple confirmed the news to People.
Gruffudd, who starred in The Fantastic Four movie, and his wife are said to be "thrilled" at expanding to their family.
The couple are already parents to three-year-old daughter, Ella Betsi.
The 39-year-old Hollywood actor, who is originally from Wales, has previously joked that his daughter keeps him on is toes.
He said: "She's very, very strong. She's like her mother. I've got my work cut out for me at home."
In the interview The Rachael Ray Show last year, the actor added that he loved spending more time with his daughter, as his partner was away filming.
He explained: "I had Ella the entire time, playing 'super dad'. It's very exciting. I love it. I can't get over what great company [a child] can be."
The couple met on the set of Disney's 101 Dalmations sequel before they tied the knot in September 2007 in a private ceremony in Mexico. They welcomed their first daughter together two years later.
