Following earlier reports on Wednesday morning, the couple confirmed the news via Twitter.
Lineker, 55, took to the social media site to say: "Thank you all for your kind words. @DanielleBux and I had many wonderful years together. We remain very close and the greatest of friends."
His wife retweeted his message, adding: "We have shared the most incredible 8 years that I will always cherish. We will remain the best of friends."
A reason for the separation has not been given, but some reports have suggested that Danielle wanted more children, while Lineker did not.
Danielle, 36, is a mum of a teenage daughter from a previous relationship, while Lineker is a father of four from his former marriage to Michelle. The couple divorced in 2006.
In 2007, he was set up on a blind date with Danielle. They tied the knot in Italy in 2009.
