The British driver was invited to join all the other drivers, along with celebrities and racing legends, at the event on Wednesday ahead of this weekend's Grand Prix at Silverstone.
However, instead he chose to go on a two-day holiday in Greece.
On twitter, Hamilton said: "To my loving fans, I can't wait to see you in Silverstone. Until then, I'm away on a two day break. God Bless you all. Love, Lewis."
However, some fans took to twitter to voice their disappointment.
One user said: "Im a massive Lewis Hamilton fan, BUT when the #BritishGP needed him the most he didn't turn up to #F1LiveLondon he's let us down massively."
And another tweeted: "Lewis Hamilton not bothering to show up to #F1LIVELONDON shows a sickening lack of respect for both the sport and its' fans."
The London event was put on by Liberty Media, the company which bought Formula 1's commercial rights earlier this year.
